Secreted, receptor-associated BMP regulators reduce stochastic noise intrinsic to many extracellular morphogen distributions

چکیده

1 pMad Local Model In our analysis, transmission of BMP signal from membrane to nucleus through Smad phosphorylation is approximated by Eq. 1. More detailed networks on nucleocytoplasmic shuttling of Smad and their downstream interpretations are given in (1). The Smad network model we consider here for signal transduction is represented using a 1 order differential equation, that includes phosphorylation of Smad growth factors and a decay term for dephosphorylation and ubiquitination of the phosphorylated Mad. For simplicity, we approximate the system with a simple linear model and more realistic networks will be analyzed elsewhere.

Secreted, receptor-associated bone morphogenetic protein regulators reduce stochastic noise intrinsic to many extracellular morphogen distributions.

Morphogens are secreted molecules that specify cell-fate organization in developing tissues. Patterns of gene expression or signalling immediately downstream of many morphogens such as the bone morphogenetic protein (BMP) decapentaplegic (Dpp) are highly reproducible and robust to perturbations. This contrasts starkly with our expectation of a noisy interpretation that would arise out of the ex...

Cell Biology Modeling Development

The fundamental problem of pattern formation for example, how to specify body axes, limbs, digits during development comes down to interpreting a common set of genetic instructions differently at different locations in space (Wolpert 1969). (See Figure 1.1 for an illustration). In all multicellular animals, this process is orchestrated by morphogens, molecules that are produced at discrete site...

Kinetics of receptor occupancy during morphogen gradient formation.

During embryogenesis, sheets of cells are patterned by concentration profiles of morphogens, molecules that act as dose-dependent regulators of gene expression and cell differentiation. Concentration profiles of morphogens can be formed by a source-sink mechanism, whereby an extracellular protein is secreted from a localized source, diffuses through the tissue and binds to cell surface receptor...

Intrinsic Noise Profoundly Alters the Dynamics and Steady State of Morphogen-Controlled Bistable Genetic Switches

During tissue development, patterns of gene expression determine the spatial arrangement of cell types. In many cases, gradients of secreted signalling molecules-morphogens-guide this process by controlling downstream transcriptional networks. A mechanism commonly used in these networks to convert the continuous information provided by the gradient into discrete transitions between adjacent cel...

Semi-adaptive response and noise attenuation in bone morphogenetic protein signalling.

Temporal dynamics of morphogen-driven signalling events are critical for proper embryonic development. During development, cells translate extracellular bone morphogenetic protein (BMP) gradients, often subject to noise, into graded intracellular tail-phosphorylated SMAD (TP-SMAD) levels. Using modelling and experimental approaches, we found that BMPs induce TP-SMAD responses in neural precurso...
